I would like to test the Hyd. pressure on my tractor. I just bought a gauge that reads up to 3000psi, a 12" hose and have ordered the proper quick coupler nipple that fits my SCV.
Can I just plug it in one of the loader QD's and move the lever to get a accurate reading? Will that tell me the relief valve pressure?

Yes and yes. Be advised that some tractors have separate relief valves for the main system and another for the loader. My TC33D is supposed to be 2130 PSI for the system and only 1800 for the loader.
