Hello new to this site, I am replacing rebuilding a 2T80 yanmar. did not mark pistons previous to removal. does it matter the direction of install?

On most engines yes. The wrist pin is off set just a tiny bit to dampen piston slap. A John Deere service manual should tell you how to figure it out. Some online are available a down loads so you wouldn't have to wait very long for the manual.
